Judul ArtikelAKTIVITAS FRAKSI ETIL ASETAT DAUN SRIKAYA (Annona squamosa Linn.) TERHADAP KADAR ASAM URAT PADA MENCIT YANG DIINDUKSI POTASIUM OKSONAT
AbstrakABSTRAK Latar Belakang: Hiperurisemia merupakan keadaan dimana terjadi peningkatan kadar asam urat diatas normal. Peningkatan kadar asam urat dapat disebabkan oleh peningkatan produksi asam urat maupun penurunan ekskresi asam urat. Daun srikaya (Annona squamosa L.) telah terbukti dapat menurunkan kadar asam urat. Tujuan dari penelitian ini untuk mengetahui aktivitas fraksi etil asetat (FEA) daun srikaya (A. squamosa L.) serta pengaruh peningkatan dosis FEA daun srikaya (A. squamosa L.) terhadap peningkatan aktivitas hipourisemia pada mencit yang diinduksi potasium oksonat. Metode: Penelitian ini merupakan penelitian eksperimental dengan Post Test Only Controlled Group Design menggunakan mencit yang terbagi dalam 5 kelompok dengan Rancangan Acak Lengkap (RAL). Kelompok tersebut diberikan perlakuan selama 7 hari dengan pemberian aquades pada kontrol negatif, Allopurinol 10 mg/kgBB pada kontrol positif, dan FEA daun srikaya (A. squamosa L.) dosis 94,5; 189; dan 378 mg/kgBB. Pada hari ke-8 semua kelompok diinduksi dengan potasium oksonat 250 mg/kgBB, kemudian diberikan perlakuan dan dilakukan pengambilan darah. Darah yang didapatkan kemudian direaksikan dengan pereaksi kit TBHBA dan diukur kadar asam uratnya menggunakan spektrofotometer UV Visibel. Hasil kadar asam urat dianalisis menggunakan uji statisika Kruskal Wallis dengan taraf kepercayaan 95%. Hasil: Rata-rata kadar asam urat pada pemberian FEA daun srikaya (A. squamosa L.) dosis 94,5; 189; dan 378 mg/kgBB berturut-turut adalah 2,127 ± 0,876; 2,471 ± 0,846 dan 2,027 ± 0,426 mg/dl. Hasil rata-rata kadar asam urat tersebut menunjukkan bahwa peningkatan dosis FEA daun srikaya (A. squamosa L.) tidak memberikan peningkatan aktivitas hipourisemia pada mencit yang diinduksi potasium oksonat. Kesimpulan: FEA daun srikaya (A. squamosa L.) belum memberikan aktivitas hipourisemia pada mencit yang diinduksi potasium oksonat.
Abstrak (Inggris)ABSTRACT Background: Hyperuricemia is an abnormally high level of uric acid in the blood. Increasing of uric acid levels can be caused by the increasing of uric acid production and decreasing of uric acid excretion. The leaves of custard apple (Annona squamosa L.) can reduced uric acid levels. The aim of this study is to determine the activity of ethyl acetate fraction (FEA) custard apple leaf (A. squamosa L.) and the effect of increasing the dose of FEA in custard apple leaf (A. squamosa L.) to increasing of hypouricemic activity in mice with potassium oxonate-induced. Method: This was an experimental study with Post Test Only Controlled Group Design used mice were divided into 5 groups with Completely Randomized Design (CRD). The group given treatment for 7 day with aquadest in negative control, Allopurinol 10 mg/kg in positive control, and FEA of custard apple leaves (A. squamosa L.) with the dose 94,5; 189; and 378 mg/kg. On the 8th day of the treatment, all groups were induced with potassium oxonate 250 mg/kg and then the blood of the sample was taken after the treatment. The blood was reacted with TBHBA of reagent kit and measured the uric acid level using Visible UV spectrophotometer. The results of uric acid levels were analyzed using Kruskal Wallis statistic test with 95% confidence level. Result: The average of uric acid level in FEA of custard apple leaves (A. squamosa L.) with the dose 94,5; 189; and 378 mg/kg continuity is 2,127 ± 0.876; 2,471 ± 0,846 and 2,027 ± 0,426 mg/dl. Thus, the conclution is the uric acid level shows that the increasing of FEA of custard apple leaf (A. squamosa L.) dose did not present an activity of hypouricemic in mice with potassium oxonate-induced. Conclusion: FEA of custard apple leaf (A. squamosa L.) had not given the activity of hypouricemic in mice which were induced with potassium oxonate.
